B-Zell-Reifungsantigen (BCMA), ein Ziel für das Multiple Myelom

BCMA wird selektiv während der Plasmazelldifferenzierung gebildet und ist auf naiven und Gedächtnis-B-Zellen fast nicht vorhanden^[1,2]. Die Wechselwirkung zwischen BCMA und seinem Liganden BAFF und APRIL  ist für die Aufrechterhaltung langlebiger Plasmazellen von Bedeutung^[3,4]. BCMA ist ein ideales Ziel für Krebserkrankungen der Plasmazellen, da es in anderen wichtigen Organen nicht exprimiert wird. Dutzende von Therapien, die auf BCMA abzielen, darunter chimäre Antigenrezeptor-T- (CAR-T) Zellen, Antikörper-Wirkstoff-Konjugate und biospezifische Antikörper, werden derzeit entwickelt und klinisch getestet. An der Spitze steht das Produkt bb2121 von Bluebird und Celgene. Eine multizentrische klinische Studie der Phase I hat eine erstaunliche Ansprechrate von 80% bei Patienten mit multiplem Myelom ergeben. Wenn sich diese Ansätze erfolgreich etablieren, werden sie Patienten mit multiplem Myelom eine dringend benötigte Option jenseits von Daratumumab (anti-CD38 Antikörper) und Elotuzumab (anti-SLAMF7) bieten.

Die Wechselwirkungen zwischen BAFF / APRIL und ihren Rezeptoren sind selektiv. Hinweis: BCMA kann sowohl mit BAFF als auch mit APRIL zusammenwirken^[5]. Die Affinität der APRIL-BCMA-Interaktion ist jedoch viel höher als die der BAFF-BCMA-Bindung^[6].

Daten zur Produktvalidierung

293 cells were transfected with anti-BCMA-scFv and RFP tag. 2e5 of the cells were stained with B. FITC-labeled human BCMA / TNFRSF17 protein, Fc tag (Cat. No. BCA-HF254, 3 μg/ml) and C. FITC-labeled protein control. A. Non-transfected 293 cells and C. FITC-labeled protein control were used as negative control. RFP was used to evaluate CAR (anti-BCMA-scFv) expression and FITC was used to evaluate the binding activity of FITC-labeled human BCMA / TNFRSF17 protein, Fc tag (Cat. No. BCA-HF254).

Protocol

Human T cells were transfected with anti-BCMA CAR and cultured for 3 days. Three days post-transfection, 1e6 cells were first incubated with 50ul Biotinylated Human BCMA, Fc,Avitag (Cat. No. BC7-H82F0, 8 μg/ml ), washed and then stained with PE Streptavidin and analyzed by flow cytometry. (Data are kindly provided by PREGENE Biopharma).

Protocol

We offer a variety of ready-to-use biotinylated proteins using our in-house developed labeling techniques, which confer high bioactivity and minimal batch-to-batch variation. In addition, we also offer FITC-labeled BCMA and BAFF proteins.
