IgG-Isotypkontrollen
Antikörper-Medikamente sind zu einem wichtigen Bereich in der Entwicklung der Biomedizin geworden. Isotypkontroll-Antikörper in der Entwicklung von Antikörper-Medikamenten gehören zur gleichen Klasse und zum gleichen Subtyp wie der gegen das Zielmolekül entwickelte primäre Antikörper, binden aber nicht spezifisch an ein bekanntes oder potenzielles Zielmolekül. Mit der Isotypkontrolle soll bestätigt werden, dass die Bindung oder Wirkung des primären Antikörpers spezifisch ist und unspezifische Wechselwirkungen mit Fc-Rezeptorproteinen oder mit anderen Proteinen ausgeschlossen sind. Inzwischen kann die Isotypkontrolle auch als Antikörper verwendet werden, der um die Bindung an Fc-Rezeptorproteine konkurriert, was mit funktionellen blockierenden Antikörpern verwandt ist, die die gleiche Funktion erfüllen, was eine effektive und angemessene Bewertung der Wirksamkeit von Antikörper-Medikamenten erleichtert. Der unspezifische Hintergrund ist eine wichtige Variable, die kontrolliert werden muss. Die Einstellung des entsprechenden Isotypkontroll-Antikörpers zum Nachweis unspezifischer Bindungen kann die Entstehung falsch positiver Ergebnisse verringern und die möglichen Einflussfaktoren im Prozess der Arzneimittelentwicklung genau bewerten.

No. DNP-PM1) and positive control (Cat. No. FM3-HPY53) respectively, and PE signal was used to evaluate the binding activity (QC tested).
