Einführung
Star Staining fluoreszenzmarkierte Produkte werden mit einer ortsspezifischen Markierungstechnologie der neuen Generation in „Star Standard“ - Qualität entwickelt. Diese Produkte wurden speziell für den Nachweis und die Überwachung von CAR-T-Zellen in klinischen Studien entwickelt.
Ein fluoreszenzmarkiertes Protein ist ein leistungsfähiges Hilfsmittel zum Nachweis der CAR-Expression in Forschungs- und klinischen Proben durch Durchflusszytometrie. Diese Proteine sind vormarkierte Fluoreszenzfarbstoffe, die die CAR-Expression durch schrittweise Färbung mit minimalem Hintergrund erkennen können. Die Entwicklung qualitativ hochwertiger fluoreszenzmarkierter Proteine stellt jedoch eine große Herausforderung dar, wodurch es bisher nur wenige handelsübliche Produkte gibt.
Die Konjugationstechnik ist ein wesentlicher Engpass für die Entwicklung hochwertiger fluoreszenzmarkierter Proteine. Derzeit ist die am weitesten verbreitete Markierungstechnik auf dem Markt die traditionelle chemische Markierungsmethode. Es ist eine sehr einfache Methode, Proteine unspezifisch mit einem Fluoreszenzfarbstoff kovalent zu markieren. Eine zufällige Modifikation kann jedoch zu Heterogenität führen und die Bioaktivität des Proteine durch Blockieren der proteinaktiven Stelle beeinflussen. Studien weisen darauf hin, dass die ortsspezifische Markierungstechnologie der neuen Generation fluoreszierende Farbstoffe ortsspezifisch an spezifische Proteine binden kann. Damit wird die Einheitlichkeit der fluoreszenzmarkierten Produkte garantiert und eine Beeinträchtigung der Proteinaktivität vermieden. Diese Eigenschaft macht diese Technologie zu einer wertvollen Komponente bei der Entwicklung hochwertiger fluoreszenzmarkierter Proteinprodukte mit hoher Spezifität und Empfindlichkeit.

FITC signal was used to evaluate the expression of CD3+ T cells in non-transduced PBMCs, and PE signal was used to evaluate the non-specific binding activity to non-transduced PBMCs.
