    PE-Labeled Human SIRP alpha, Fc Tag (Cat. No. SIA-HP252) is produced via site-specific conjugation of PE to Human SIRP alpha, Fc Tag under optimal conditions with a proprietary technology. Human SIRP alpha, Fc Tag is expressed from human 293 cells (HEK293). It contains AA Glu 31 - Arg 370 (Accession # P78324-1).

    FACS analysis shows that the binding of PE-Labeled Human SIRP alpha, Fc Tag (Cat. No. SIA-HP252) to Jurkat cells overexpressing CD47 was inhibited by increasing concentration of neutralizing anti-human CD47 antibody. The IC50 is 0.37 μg/mL (QC tested).

    I recently used the PE-Labeled Human SIRP alpha / CD172a Protein, Fc Tag for detecting the expression of CD47 in Jurkat T-cells and primary T-cells, and I am very pleased with the results. The SIRP alpha reagent effectively binds to CD47 overexpressed in both Jurkat and primary T-cells. This experiment was conducted to demonstrate the overexpression of CD47 in T-cells, using the anti-CD47 antibody (clone: CC2C6) to confirm the expression. Additionally, it was crucial to show that our CD47 can bind to its natural ligand, SIRP alpha. The reagent performed excellently in this regard, providing clear and reliable results. I've attached an image showing the data with a CD47 KO Jurkat cell line, further validating the specificity and effectiveness of this reagent. Overall, I highly recommend this product for CD47 detection and binding studies.

    I used this SIRPa-PE Fc tag for a FACS assay using the recommended SOP provided by Acro and our rep Nik Kamachi. Nik was very helpful in providing insight on the capabilities of Acro to provide custom labeled SIRPa Fc tags to meet our needs for small molecule screening assays that are FACS and IF-based. The human SIRPa-PE Fc tag was very easy to use and gave a strong signal with both flow cytometry and imaging

Background Introduction

Tyrosine-protein phosphatase non-receptor type substrate 1 (SHPS1) is also known as CD172 antigen-like family member A (CD172a), Macrophage fusion receptor, MyD-1 antigen, Signal-regulatory protein alpha (SIRPA or SIRP alpha) or p84, is a member of the SIRP family, and also belongs to the immunoglobulin superfamily. SIRP alpha is Ubiquitous and highly expressed in brain. SIRPA / CD172a is immunoglobulin-like cell surface receptor for CD47 and acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. SIRPA / SHPS-1 supports adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment and may play a key role in intracellular signaling during synaptogenesis and in synaptic function By similarity. SIRPA / MyD1 involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors or insulin and mediates negative regulation of phagocytosis, mast cell activation and dendritic cell activation. CD47 binding prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.
