Source
PE-Labeled Human Siglec-3, His Tag (CD3-HP2E3) is produced via site-specific conjugation of PE to Human Siglec-3, His Tag under optimal conditions with a proprietary technology. Human Siglec-3, His Tag is expressed from human 293 cells (HEK293). It contains AA Asp 18 - His 259 (Accession # AAH28152.1).
Predicted N-terminus: Asp 18

5e5 of anti-Siglec-3 CAR-293 cells were stained with 100 μL of 1:50 dilution (2 μL stock solution in 100 μL FACS buffer) of PE-Labeled Human Siglec-3, His Tag (Cat. No. CD3-HP2E3) and negative control protein respectively. PE signal was used to evaluate the binding activity (QC tested).

We purchased this reagent for use in detecting CD33 CAR expression and it worked as advertised. The PE signal gave great separation by flow cytometry and allowed for the clear detection of CAR expression on the surface of our cells. The product page states that the CD33 PE reagent is incompatible with streptavidin-biotin detection systems. When we tried to co-stain with another biotinylated reagent, subsequent application of streptavidin-APC resulted in high non-specific APC staining. This could be avoided by staining with CD33 PE after all other staining steps have been completed.

We purchased this antibody to use for flow cytometry on a client study, who specifically requested Acro antibodies! They are reliable and have continued to be one of our top providers of antibodies. We typically use multiple fluorescent antibodies with various protein targets and fluorophores at once for flow cytometry panels in human and mouse tissues, and Acro antibodies have always given us a great readout.
