FACS Analysis of Blood Samples

After Fc receptor blockade, whole blood only control (Figure A) and whole blood spiked with CAR-T cells (Figure B) were first stained with PE-Labeled Human CD19 (20-291) Protein, His Tag (Cat. CD9-HP2H3), following 7-AAD staining. The data were analyzed using FCS Express 7 software (Routinely tested). CAR-T cells were identified as PE-positive events within the live CD45+CD3+ (or CD45+) lymphocyte population. The proportion of CD45+CAR+ cells is around 20%, which is largely consistent with the theoretical spike-in value.

Bioactivity-FACS

1e6 of the anti-CD19 CAR-293 cells were stained with 100 μL of 1:50 dilution (2 μL stock solution in 100 μL FACS buffer) of PE-Labeled Human CD19 (20-291), His Tag (Cat. No. CD9-HP2H3) (QC tested).

We use this reagent to detect CD19 CAR on the cell surface by flow cytometry. We use it at the recommended dose and are very happy with the results.

I run lentiviral titers at work and desperately needed a CD19 antibody due backorder. I stumbled upon Acro Bio's PE-CD19 abs and it worked wonders! When I first used it, there was higher CD19 expression than I typically see in my vectors. I performed a comparison study testing out various CD19 (some conjugated some not conjugated) antibodies. The Acro Bio antibody had the best performance! It showed greater binding than the usual CD19 antibody I use. After I conducted this study, my company set their heart's desire to Acro's CD19 antibody. We solely use this antibody for all our CD19 projects. AND its more affordable than many other CD19 antibodies on the market! This is such a phenomenal antibody! I highly recommend it!

Thank you so much ACRO for your CD19 materials. I work in a CAR focused lab and the CD19 we get from you is instrumental to our operations. I work in immunology and this antibody is used in all my downstream flow assays to check the status of our novel CAR materials. I use it at least weekly, and the staining is easy and immaculate. My peaks are clear and I couldn't be happier.
