2e5 of Anti-CD19 CAR-293 cells were stained with 100 μL of 1:50 dilution (2 μL stock solution in 100 μL FACS buffer) FITC-Labeled Monoclonal Anti-FMC63 Antibody, Mouse IgG1 (Y45) (Cat. No. FM3-FY45) and isotype control respectively. FITC signal was used to evaluate the binding activity (QC tested).
Protocol
Non-specificity of FITC-Labeled Monoclonal Anti-FMC63 Antibody, Mouse IgG1 (Y45) (Cat. No. FM3-FY45) binding to CD3+ cells present in human PBMC. Human PBMCs were simultaneously stained with PE-labeled anti-CD3 antibody and FITC-Labeled Monoclonal Anti-FMC63 Antibody, Mouse IgG1 (Y45) (2 μL of the antibody stock solution corresponds to labeling of 5e5 cells in a final volume of 100 µL), washed and then analyzed with FACS. Both PE and FITC positive signals was used to evaluate the non-specific binding activity to human CD3+ cells (QC tested).

My team purchased the FITC-conjugated Anti-FMC63 antibody (FM3-FY45) to characterize a CAR-T cell line that we had developed. We used the antibody in our flow cytometry experiment, and we saw virtually no background signal in our negative control samples (0% and 1% FITC readout). We did, however, get beautiful results for our CAR-T cell line samples and were able to confirm successful transduction. The instructions for use in the CoA were very clear, and the recommended concentration was sufficient for our purposes. I am very interested in revisiting ACRO Biosystems for more antibody purchases in the future.

We purchase the anti FMC63 to detect Chimeric antigen receptor (CAR) cells in patients treated with an academic CAR for lymphoma. We used the antibody to detect by flow cytometry the cells that express the CAR and the persistence of our modified T cells. We are extremely satisfied with the performance of the antibodies. We are developing new CAR and for sure we will use the antibodies from Acrobiosystems.

We are a CAR-T therapy research group and we use FITC anti-FMC63 and FITC-Labeled Human CD19 protein to detect CAR19 cells both in patient blood and in vitro cultures by flow cytometry. The results obtained so far with both reagents are reliable and reproducible, with a clear and easy detection of positive population even when reagents are directly incubated with blood samples.
