CD45 Background

CD45 is a receptor protein tyrosine phosphatase, also known as Ly-5 or leukocyte common antigen. CD45 mainly involves in the initiation of T cell receptor signaling by controlling the activation of the Src family protein-tyrosine kinases Lck and Fyn. CD45 deficiency causes in T- and B-lymphocyte dysfunction in the form of severe combined immune deficiency. It also takes a significant role in autoimmune diseases and cancer as well as in infectious diseases including fungal infections.

The biotinylated protein was immobilized on the SA biosensor of Octet BLI R8 to measure the binding affinity of anti-human CD45RA antibody. The assay worked on the first try!

    We were very satisfied with the whole process of purchasing and utilizing the product as we had no issues with shipping time or product packaging. We used the protein to assist in the development of LFAs detecting for CD45 on cells. Pairs of anti-CD45 antibodies were first tested using the protein to help us determine which pair of antibodies produced the best results. These tests allowed us to move forward with designing full LFAs.

Protocol for QC was easy to understand and repeat. Easy way to download the Certificate of Analysis via the printed QR code on product vials. Customer service answered very fast and was very friendly. QC of biotinylated human CD45 protein vs. Human Galectin-1 protein (GA1-H4113) was fine. Biotinylated human CD45 protein worked fine and could be loaded on Streptavidin biosensors (Octet system).
