    Her2 PRODUCT SHOW

    This product can be directly isolated using conventional column-free magnetic stand (eg. Cat. MB-01/ MB-02/ MB-03/ MB-04). Please note that due to its micron-scale size, it is not compatible with commercially available column-based (M/D/L) magnetic separation systems.

  • Product Description

    ActiveMax® Human Her2 μBeads, premium grade (for cells) are uniform, superparamagnetic beads of 5.5 µm in diameter immobilized with Human Her2 protein expressed from human 293 cells (HEK293) and contains AA Thr 23 - Thr 652 (Accession # P04626-1).
    ActiveMax® Human Her2 μBeads, premium grade (for cells) are produced under sterile manufacturing conditions (ISO 5), and no animal- or human-derived components are used throughout the production process. It is produced under our rigorous quality control system that includes a comprehensive set of tests including sterility and endotoxin tests.

  • Application

    ActiveMax® Human Her2 μBeads, premium grade (for cells) are designed to stimulate in vitro Her2-specific CAR-T cells or UCAR-T cells, similar to the tumor cell lines that express human Her2 antigen. It can be used as follows:
    Evaluating the characteristics of CAR-T cells or UCAR-T cells.
    In vitro expansion of Her2-specific CAR-T cells or UCAR-T cells.
    In vitro enrichment of Her2-specific CAR-T cells or UCAR-T cells.

  • Reconstitution

    See Certificate of Analysis (CoA) for detailed instruction.

  • Storage

    This product is stable in storage under the following conditions: -20˚C for 12 months in lyophilized state. -70°C for 3 months under sterile conditions after reconstitution.

    Please protect from light and avoid repeated freeze-thaw cycles.Immediate use after reconstitution is highly recommended.

    Assay of Human Her2 on the μBeads surface by Flow cytometry. ActiveMax® Human Her2 μBeads, premium grade (for cells) (Cat. No. MBS-C006) surface was fluorescently stained using Anti-Her2 antibody and PE-anti-Human IgG Fc, analyzed by flow cytometry (QC tested).

     Her2 CYTOKINES

    ActiveMax® Human Her2 μBeads, premium grade (for cells) (Cat. No. MBS-C006) can activate Her2-specific CAR-T cells by detecting the secretion of IFN- γ in vitro.

     Her2 CYTOKINES

    HER2 CAR-T cells were stimulated with ActiveMax® Human HER2 μBeads (Cat. No. MBS-C006) for 24h, the cell-free supernatant was collected by centrifugation for evaluating IFN-γ, TNFα and IL-4 secretion by CBA. The results indicated that HER2 CAR-T cells responded to HER2 μBeads, and the expressions of the IFN-γ, TNFα and IL-4 increased (Data is the average of 3 independent experiments, error bars ± SD).

  • Stimulate Her2 CAR-T Cells -Cytotoxic Molecules

     Her2 CYTOTOXIC MOLECULES

    HER2 CAR-T cells were stimulated with ActiveMax® Human HER2 μBeads (Cat. No. MBS-C006) for 24h , the cell-free supernatant was collected by centrifugation for evaluating Granzyme B secretion by ELISA. The results indicated that HER2 CAR-T cells responded to HER2 μBeads, and the expressions of the Granzyme B increased (Data is the average of 3 independent experiments, error bars ± SD).

  • Stimulate Her2 CAR-T Cells -Activation Marker

     Her2 ACTIVATION MARKER

    HER2 CAR-T cells were stimulated with ActiveMax® Human HER2 μBeads (Cat. No. MBS-C006) for 24h. After demagnetization of the beads, the cells were collected and CD25, CD69, and CD137 were detected by FACS. The results indicated that HER2 CAR-T cells responded to HER2 μBeads, and the expressions of the three markers increased (Data is the average of 3 independent experiments, error bars ± SD).

  • Stimulate Her2 CAR-T Cells -Exhaustion Marker

     Her2 EXHAUSTION MARKER

    HER2 CAR-T cells were stimulated with ActiveMax® Human HER2 μBeads (Cat. No. MBS-C006) for 24h. After demagnetization of the beads, the cells were collected and LAG3, PD-1 were detected by FACS. The results indicated that HER2 CAR-T cells responded to HER2 μBeads, and the expressions of the Granzyme B increased (Data is the average of 3 independent experiments, error bars ± SD).
