  • Background
    ClinMax™ ELISA Kit is convenient ready-to-use immunoassay Kit, specifically designed to quantitate human GM-CSF that is present in complex biological samples, such as human serum, plasma, and cell culture supernates.

    A comprehensive validation of the ELISA method was performed following the ICH M10 on bioanalytical method validation and the FDA’s bioanalytical method validation guidance for industry. This validation included assessments of linearity, accuracy, precision, dilution linearity, recovery, and the hook effect. For details information, please refer to the DS.

  • Application

    The kit is developed for quantitative detection of GM-CSF in human serum and cell culture supernates.

  • Reconstitution
    Please see Certificate of Analysis for details of reconstitution instruction and specific concentration.
  • Storage
    Unopened kit should be stored at 2℃-8℃ upon receiving. The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.
  • Assay Principles
    This assay kit is used to measure the concentrations of human Granulozyten-Makrophagen-Kolonie-stimulierende Faktor (GM-CSF) by employing a standard sandwich-ELISA format. The microplate in the kit has been pre-coated with Anti- GM-CSF Antibody. First add the standard samples provided in kit and your samples to the plate, next add detection antibody Biotin-Anti-GM-CSF Antibody to the plate, incubate and wash the wells. After wash add HRP-Streptavidin to the plate, incubate and wash the wells. Lastly load the substrate into the wells and color develops in proportion to the amount of Interferon-γ bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm and 630nm.

For each experiment, a standard curve needs to be set for each microplate, and the specific OD value may vary depending on different laboratories, testers, or equipment. The following example data is for reference only. The sample concentration was calculated based on the results of the standard curve. The minimum detectable concentration of GM-CSF is less than 10.0 pg/mL.

Validation
Intra-Assay Statistics

Ten replicates of each of 3 samples containing different GM-CSF concentrations were tested in one assay. Acceptable criteria: CV<10%.

Three samples containing different concentrations of GM-CSF were tested in independent assays. Acceptable criteria: CV<15%.

Recovery

Recombinant GM-CSF was spiked into 5 human serum samples, and then analyzed. The average recovery of GM-CSF for serum samples is 102.82%.

  • Background: GM-CSF
    GM-CSF, Granulocyte Macrophage Colony Stimulating Factor
    GM-CSF is a colony-stimulating factor that regulates growth and differentiation of hematopoietic cells. GM-CSF is a primarily T cell-derived 14-35 kDa variably glycosylated protein which affects growth and functions of granulocytes and cells of the macrophage lineage at all stages of maturation.
    It is also required for the development of specialized alveolar macrophages responsible for pathogen surveillance and the metabolism of pulmonary surfactant. Its production is widespread throughout the body in certain infectious and inflammatory states to enhance a local innate immune response. On the other hand, its abnormal production can lead to chronic inflammatory states as observed in rheumatoid arthritis and asthma. Some of the actions of GM-CSF include enhancing macrophage, neutrophil, eosinophil, and natural killer cell effector functions; dendritic cell survival and differentiation; and the up-regulation of antigen presentation.
